Exasmosis in a living cell when placed in a hypertonic solution, is called plasmolysis.
Aim To study plasmolysis in epidermal cells of Tradescantia leaf.

Procedure:
Take a fresh Tradescantia leaf and peel off its lower-colored surface so that epidermal cells can be observed.
Take a small piece of epidermal peel and place it in a concentrated sugar solution for a few minutes (hypertonic solution). Mount the slide and put a coverslip.
Observe the slide under a microscope and focus on a few cells.
Take another small piece of peel from the lower epidermis and place it in water (hypotonic solution) for a few minutes, mount it on a slide, and put a coverslip.
Carefully observe this slide also under the microscope.
Observation:
The slide prepared using hypertonic solution shows shrinkage of cytoplasm (protoplast) cytoplasm moves (recedes) away from the cell wall and becomes concentrated in the corner of the cell. This is known as plasmolysis.
The slide prepared using a hypotonic solution or water shows that cell cytoplasm bulges or swells i.e. increase in size. This is due to endosmosis Inferences:
